What Is Kisspeptin‑10?

Kisspeptin‑10 is a synthetic decapeptide derived from the endogenous Kisspeptin family, originally identified as metastin from the KISS1 gene. It is a potent activator of the G‑protein‑coupled receptor GPR54 (also known as KISS1R), which plays a central role in reproductive hormone regulation. Kisspeptin‑10 has been studied extensively in neuroendocrinology for its ability to modulate gonadotropin‑releasing hormone (GnRH) secretion, thereby influencing fertility, puberty, and reproductive health.

Mechanism of Action

Kisspeptin‑10 binds to KISS1R receptors located on hypothalamic GnRH neurons. This receptor activation triggers intracellular signaling cascades involving phospholipase C and calcium mobilization, leading to pulsatile GnRH release. Elevated GnRH subsequently stimulates the anterior pituitary to secrete luteinizing hormone (LH) and follicle‑stimulating hormone (FSH), which regulate gonadal steroidogenesis and gametogenesis. Kisspeptin‑10 is considered a master regulator of the hypothalamic‑pituitary‑gonadal (HPG) axis, with additional roles in metabolic and neuroendocrine signaling.
